Pancreatic cancer – Stemness and Resistance to Treatments such as Electroporation/Electrochemotherapy

Principal Investigator: Dr. Patrick Forde

Researcher: Muhammad Shahzad Ali

This is a collaboration project setup by Professor Giuseppe Saglio University of Turin and Dr. Patrick Forde, University College Cork.

Pancreatic cancer is one of the deadliest cancers; one-year survival in the UK is only 20% with 8,500 new cases diagnosed annually. Currently in Ireland Pancreatic cancer is limited which is amazing when pancreatic cancer (PDAC) is currently the fourth leading cause of cancer-related death and its incidence is expected to more than double by the year 2030.

The clinical usage of electroporation treatments has been on the rise. This project looks at current treatments and electroporation delivered drugs. From our interpretation of the literature, there has not been a study looking at the effect of stemness and resistance to electroporation delivered treatments.

The project will evaluate the stemness factors (example: Nanog, Sox4, Lin28, Oct4) relating to the treatment and determine the change in stemness factors. The presence of these markers and their respective expression can relate to the resistance of pancreatic cancer to the treatment regime been evaluated. Once stemness markers are assessed the project intends to determine the relevance of the respective pathways as therapeutic targets and measure the role of the lysosome in electroporation treatments. The project has shown that electroporation can modulate the workings of lysosomal activity and we believe the lysosome has a role in DAMP protein release. The group is working on immunogenic cell death through DAMP protein release. Therefore, the project intends to link and evaluate electroporation modulation of lysosome and its role in immunogenic cell death. This is a potential therapeutic target
